I am new to reloading so bare with me. I have a lee 3 die set for .45 acp. Do I have to use the sizer die with new unfired brass? Shouldn't it already be sized?

Yes, it should be sized.
But I always size & neck expand new brass anyway just to insure proper bullet pull that I know is going to be right.
It is often-times bent or dinged on the mouth from packing & transport.

I'll supplement what RC said.
If your loading shouldered rounds you should resize to get a consistent headspace, or adjust the die so all rounds will be. Same thing applies to non-shouldered rounds, you need a good starting point.
Accuracy's starting point is consistency.

yep past few times when i bought new brass. If you look really hard it was usually dented or just plain out of round on the end usually due to hitting other cases. so i full size all the time on new brass

I always resize plus it gives you one more look at the brass. If you always do it you will never have to worry if it's been done.
