For me with a Single-Six, focusing on follow through tightens my groups. Keep that front sight on the bull even after you fire.

No one has mentioned the 'ball and dummy' drill, mixing some spent ones with the live ones, give the cylinder a blind spin, and blaze away. Even better, let someone else load the gun.
Whatever sins you may be committing when pulling the trigger will become entirely evident when the gun doesn't fire.

White-out is OK. It's sort of a "latex" . Fingernail polish is available in white, and it also helps act as a primer, if you want to paint your sights green, orange, or red. Fingernail polish is also far more durable than White-out.
