From a legal standpoint, you need to know the 4473 up and down. Also the process of NICS checks, and that is the minimum. Add to that whatever your local/state laws have, then whatever the store's policies are, inventory control, safety, etc. From a business standpoint, you need to know the products, and be incredibly patient with people's opinions, intelligence (or lack thereof), and every other annoying thing that customers may throw at you.

The store's careers page says you have to pass a Form 4473 Background Check for some locations, but not all. Managers(where it says, "Knowledge of Guns and Ammo preferred.", not required.) always have higher requirements that don't apply to other employees. Otherwise, working in a gun shop isn't as glamorous as it appears. Usually minimum wage with no benefits and weekends and evenings for part timers.
"...let me do the gun counter..." Read the job description. Usually has, "and other duties as assigned." in the fine print.

Almost all the legal issues that come up during a sale are answered right on the 4473 itself, usually in the instructions on the back pages. All you have to do is read it, or have the customer read it (depending on the question).

Your customer has multiple residences and wants to know which one to put on the 4473? Read the instructions for question 2 on page 4.

You customer doesn’t have their actual address on their ID but has a P.O. box listed instead? It says right on the front of the form that you can’t use a P.O. box.

Your customer doesn’t have their actual current residence listed on their ID and wants to know what extra documents they need to prove residency? It’s explained on page 5 in the instructions for question 18b.

If the customer is buying the gun for someone else and is unsure if they’re committing a straw purchase, that’s answered on page 4 in the instructions for question 11a.

A customer was convicted of a felony and served time but later had the conviction expunged, and they want to know how to answer question 11c? Tell them to read the corresponding instructions on page 4.

I’m always amazed at how many stories I hear of a dealer who gave out bad information or made mistakes regarding the rules, when all they had to do is just read the instructions on the 4473 and it would tell them the answer right then and there.
