Does the NFA actually cover anti-tank guns?

Sure. Anything over .50 caliber is a Destructive Device. But unlike machine guns, you can still make new ones.
Tires are probably a pain to source.
I'll guess you trailer it to places to go shoot. (Although it would be tempting to get a Kettenkraftrad to go with it .)
Of course, any explosive round also requires a DD tax stamp, so playing with stuff other than inert projectiles is a pretty spendy venture for someone who doesn't hold a 10/02 (and 10/02 is an expensive license to maintain).

Yes they did after these where framed useless against tanks they where relegated to infantry support
