I haven't been able to find any dedicated speedloader pouches that use a MOLLE interface. Would MOLLE frag grenade pouches work?

I'd say they're too big as they are usually designed for a M67 frag. A few yrs ago Bianchi made the UM-84 holster series for the revolver ( the UM-84R)
Bianchi may have made a speed loader case that used a similar attachment as the holster, if so, it may work well w/ MOLLE systems.

Somebody makes them, I remember seeing a shooter at a three gun match who was using a revolver and vest attached speedloader pouches.
