How does shooting on a downward or upward angle effect your point of impact?

I understand and agree that out of a perfectly level barrel it is immediately dropping out of the barrel, but I cannot think of a single instance where this would even be applicable in shooting as the barrel must have at least a slight elevation (muzzle in relation to chamber) in order to effectively use any manual sighting system that I can imagine.
 
You and I are sitting on top of a 1000 foot cliff and only have an M-79 40mm grenade launcher to drive off the hoardes approaching the ladders at the bottom of the cliff. How can we gain impacts as far from the bottom of the cliff as possible.

A 40 to 45 degree angle works on level ground,,, But we have a 1000 foot drop. Would a straight and level barrel give us the most range in this situation???
 
HA.... well.. I guess that was a situation I hadnt fathomed... dont know how i could have missed it...:D

that said, I would still be inclined to think a slightly elevated barrel would provide you with the greatest range. It would, of course require some trial and error but in your posed situation, though at a certain (unknown to me) incline you would lose some range, I cannot see a Level shot maximizing your range even in this situation.. it is only 1000 feet after all...;)
