How much would it cost to have a 45 acp cylinder made for a Colt Cowboy?

Also who can do the work? One more question is ruger still making a Vaquero in 45 lc and ACP with a spare cylinder?

I believe the Colt Cowboy used the same cylinder as the Single Action Army, and since Colt made .45 ACP cylinders for them I'd call their Customer Service Departemnt and find out if they are still available.
 
Cimarron makes a dual cylinder model too

I have one in the 1873 model (with firing pin spur) and love it.

Cheap .45 ACP to practice with and a fast switch to the .45 LC.

I'm still looking for another one in fact. Very nicely balanced and beautiful wood work on the grip.

I'm not sure, But I believe Cimarron's cylinders are the same as Colt's 1st and 2nd generation 1873 models, where the 3rd. generation Single Actions and Cowboy models have a different ratchet pattern and use a different hand. The changes were made to reduce hand fitting.
