I took my Ruger GP100 to the range the other day after not shooting it for approximately 2 years (although I dryfire/practice with it regularly). After shooting it two years ago, I cleaned and lubricated it. This time, after firing twice, the trigger would not pull back and neither would the hammer. However, by manually rotating the cylinder, I was able to cock the hammer. When I brought it home to clean it, the same problem occurred when I dryfired it. The trigger would depress about halfway and then wouldn't go any further. Same thing when I tried to cock it. After cleaning and lubricating it, it functioned flawlessly

Can this problem be simply attributed to a lack of lubrication? Is there a likely internal problem? Would sending it to a gunsmith be advisable?

When I got my GP (new) the first thing I did was play with it a lot, having heard that the action was rough out of the box. Didn't take long for it to develop a squeak on cocking, which lubrication got rid of. I suspect it was the knob on the mainspring strut rubbing hard in the dry hammer socket. Because of that I make sure that mine stays well lubed.

One thing I can recommend is to NOT follow the manual and use the mainspring strut as a takedown tool to pop out the trigger group. Putting burrs on the strut knob could cause the hammer to bind.

If the trigger (and hammer) is locked, has it been released fully? Partial release doesn't allow the parts to reset and return to their normal position of rest.

No, the trigger had been fully released. I know what you mean, but that wasn't it.
