Pietta 1851 .44 Navy revolver won't cock after re-assembly.

That just about says it. The hammaer won't even go to half cock. Do I have the cylinder lined up wrong? Wedge in too tight? Any ideas?

Take the barrel and the cylinder off the gun and try to cock it. If it won't go I would say that you will have to tear it back down and check it over again, you might have assembled it wrong. Did you get the spring on the hammer right? Might have came off the roller.
 
Before you tear it apart again give it a good rap with the heel of yer palm on the side of the cylinder. That may jar it enough to let the bolts finger snap up onto the top of the hammers cam. Sometimes a new gun does that until it breaks in some or you file the edge of the bolt finger some so it doesn't hang up just before it should snap back onto the top of the hammers cam. If the bolt finger doesn't get back on top the hammers cam then the hammer can't go backwards because the hand can't go up because the cylinder can't rotate because the bolt won't come out of the cylinder notch because the hammers cam isn't under the bolts finger to raise it so the other end of the bolt is freed from the cylinder notch. Whew...
