Is it possible to load a 12 gauge rifled slug lighter than 437 grains, for instance loading a 250 grain slug in a regular size 2 3/4 shell
This. Just use a round ball. The standard 12 gauge slug is awfully short to begin with and if you shorten it more you might as well use a round ball. Wads are no problem as the standard rs load uses card and fiber stacked to whatever length is needed for a good crimp. What are you trying to achieve and why?
