Depends on how the cartridge is going to be handled during loading into the chamber. On a single shot the bullet only has to be seated far enough to keep it from falling out of the case and enough so it just touches the rifiling when the action is closed. If the cartridge is going to have to withstand the rigors of feeding from a magazine and be subject to the recoil of other rounds, then it will need a better seat to keep from being upset. Stringers answer is a good rule of thumb.
