I picked up a used Kahr PM9.

After break in, how many round of my chosen defense round should I shoot through it without failure before considering it good to go?

At this point I just have some Federal Premium 135 Hydro-Shok Personal Defense.

After break-in I usually run a couple of mags of HP carry ammo and a couple of mixed ball and HP. If it runs okay through those I'm good to go. If I change carry ammo I do the same thing again to be safe.

trimore: I would periodically empty ur carry magazine down range for reliability and to also freshen up your carry rounds. I dont' see a need for 200 defense rounds to prove reliability either. YMMV

A couple of boxes and then half a box per month (reassures you that it functions and puts fresh ammo in the mag).

i say what ever makes you comfortable as you are the one that will be carrying it and your life will be the one on the line, however, i personally do 2-3 boxes after i get it narrowed down to the exact round that i am gonna use.

I'd fire at least 10 full magazines from each magazine you intent to carry without ANY mishaps before trusting the weapon for carry

That is a very touchy feelly question. If you want to get scientific, it takes about 300 consecutive successful repetitions of a pass fail test to be 95% certain that 99% of future repetitions will be successful. But since 300 rounds is enough to change the dynamics of a firearm, it is still worthless (or close to it). If you want a long winded run on discusion about it, I wrote a page a while back:

I personally take two mags full and dry cycle the rounds to feel for feed issues and then shoot them and call it good.

I know the Kimber hater in here don't want to hear it, I shot 100 rds of WWB, followed by 200 rds of hydra-shoks through the new gun, error free, and called it good for break-in AND carry check. When I switched to HSTs and different mags, I went through 100 of them and called it good.

At least fill and empty your carry mags of ammo a few times. As was stated above, the more you shoot, the more your firearm wears, so testing has a point of diminishing returns.
Get a bunch of ammo, get to an outdoor range, and blow up some jugs of water, you'll feel better after doing it, and should identify any feeding issues with any particular ammo.
