I'm thinking of picking up a Silencerco Octane as primarily a 300 blackout can, but also for use on pistols. A 45 would let me use it with 45s (obviously) while a 9 wouldn't. Since my primary use would be as a serviceable 300 blackout subsonic can, how much suppression effectiveness would I be sacrificing on the rifle if I went with the larger bore can?

I have this exact setup and love it. Use it for .300 in 16" barrel AR and it is nice and quiet. Shoot a lot of sub .40's through it also. A lot of people say they sound bad but i am begining to think they either have not actually tried them or the Octane is just that good of a suppressor. Highly recommend.

I'd wait for the Silencerco Hybrid if thats your goal.
I'm trying to play 'beat the clock' to a degree. I would like to get one more can in under the wire before 41P goes into effect if it does. I know we have a new CLEO sign off law in KY but I don't want to have to make my wife and BIL do fingerprint cards and pix under the proposed new rules. So I'm really looking to buy from something in stock and not even wait for a Form 3 transfer.

I run 300 BLK through my Tirant 45 sometimes. Not often though as I have a few dedicated 30 cal cans. It works. Not as quiet as the actual rifle cans by a small margin . I just don't do it often as Ive got my QD mounts for my rifles "glued" on and thats pretty much the way they stay. I don't use the TiRant much. I really need a lever action for that thing.

My last Form3 transfer was under 4 weeks.

Ive never had a dealer wait for the form 3 before filing the form 4. Not once in 20 years. Form 3's get filed at the same time as form 4's all the time. I'm at the point where if my dealer filed a form 3 and waited until it came back to file the form 4 I'd find another dealer. Despite what some dealers might claim there is no requirement for the dealer to wait for the form 3 before filing the form 4. Does is cause a little confusion? It can but the ATF doesnt seem to mind all that much. Might not be the best practice and I'm sure someone will chime in about "what if,..." but in practice it works.

My Octane 9 is quieter with 300 Blackout subs than my SDN-6. And considering the Octane 45 is quieter with 9mm than the Octane 9 is (as long as you're using the 9mm endcap), the Octane 45 with the 9mm endcap is almost certainly quieter with 300 Blackout subs than both the SDN-6 and the Octane 9.
