I just got done cleaning my GP-100 (stainless). It was a follow-up cleanig after cleaning it a week ago. I used shooters choice last week. This week I used Kleen Bore lead removing cloth, some Gun Scrub, and then oiled it down with Miltec. When I went to close the cylinder, it wouldn't close. I tried a few more times and it finally closed. Then the cylinder would bind up while I was dry-firing trying to figure out what was going on (yes, it was in a safe direction, yes, ammo was in a clear container). After a couple of minutes, it just stopped being a problem. Did my metal expand or something? What the heck was that about?

Check that the ejector rod is tight. If they get loose and start to back out then you have trouble closing the cylinder and the cylinder will tend to bind. Tighten it back up and everything works again.

Check for a small piece of patch or dirt under the extractor star.

If one of those two answers don't fix it ...... I'll be surprised. Almost gotta be down to star binding.

The ejector rod on the Ruger GP-100 cannot unscrew and it has nothing to do with locking the cylinder in place. I would bet that there is something under the extractor star.

as far as QA, my new (last year) GP-100 is awesome, there were a couple of rough spots (non-visible)...but I've never had much of a problem with it....(well, I had a high primer a while back, not the GP's fault ;-)
Against the conventional wisdom that says the Ruger is the "AK" of the D/A revolver world, both trainers and myself have noted over the years that 101's and GP 100's tend to tie up on our dirty practice ammo MUCH more often than similar S/W or Colt guns. Maybe they have tighter tolerences than Rugers given credit for 
One thing that does strike me as a possibility is that you said you just cleaned it. One time I lost a single bristle off a brass cleaning brush that worked it's way in and made the gun feel rough till I found out what was going on. Weird but true.
