I'll be a brief as I can so here goes...

1) If one calibrates ones .38 revolver for close range shooting at 21 feet, will the sights need readjustment when shooting from the 15 yard line?

2) I realise that one should practice extensively with one's choosen self preservation load (130gr .38 +Ps WWBs in my case). However if the aforementioned load is too expensive to practice with (due to import taxes and duties), how do fiocchi 158gr LRNs approximate as a training load? (one is lighter and loaded to +P while one is heavier and loaded hotter than usual) Does the recoil and point of impact differ much from a S&W K frame with 4" barrel at the 21 feet, 15 yard and 25 yard lines?

At the distances you mentioned the point-of-aim/point-of-impact will not change enough to matter. 50 meters/yards would be something else.

As for the ammunition, you'd have to try both and see what the difference in impact was from a particular revolver. I suspect it wouldn't be much at close range, but you probably would feel a difference in recoil, with the heavier bullet load having more.

My 3" revolvers are sighted in at 25 yards.
My 6" revolvers are sighted in at 50 yards.

At 25 foot self defense ranges you need to shoot fast enough that precise aiming isn't needed. I do however know where to hold for the best accuracy in case I ever have to take a precision shot at the shorter ranges.

It'a all a matter of practice.
