That's a hard question!

O/U guns were made in muzzle loading days by a number of gun makers.

And German gunmakers were building stack-barrel rifles & shotguns in the 1800's.

But perhaps the best known, "first", and best modern O/U shotgun might be the John Browning designed Superposed, introduced in 1930 and sold by FN Browning.

A lot of research in museums, etc. might find the "earliest known" over-under, but it may not be the first.

Several years ago, Beretta sold a percussion over-under replica of a mid-19th century gun of theirs.

If you want a breechloader, Greener would make you one in 1909 or earlier.

On the other hand, I have seen pictures of flintlock over- unders.
 
Well, original "over and unders" were made in the 16th century as double firing arquebuses and pistols using wheelock firing mechanisms. Very likely immediately adopted for sporting purposes by the nobility. Probably in Italy or Germany.
