Mag in, chamber empty, safety off, in case of a fire so no chance of a round cooking off. All my handguns stored in a vehicle kept this way. All truck guns are considered a backup so charging would be a minor thing IMHO. Most of the people here in this area of rural Maine do it this way. Long guns have mag out as well due to laws here.
 
Here in Colorado a long gun can not legally have a round in the chamber in a vehicle. I don't normally carry a "truck gun", but if I did it would be full mag, empty chamber, safety off.
 
I hope all of the “loaded mag empty chamber” responses are also “bolt closed”

In an ideal world I would have a side folding stock on an AR pistol. Mag in, if space allows. Chamber empty.
 
I have a H&R partner 12 gauge in the jeep. Mag is loaded with 00, chamber empty, gun is in a cheap nylon case.
 
Any gun in my truck that I have decided stays in the truck for any length of time, be it a handgun or AR, is considered a back up gun. Its nothing I plan in getting to quickly. Its an extra tool should i find myself in a situation where things are escalating (broken down car in the middle of nowhere, stuck at the cabin, etc) and need something more.

I keep the gun cased with the magazine removed and spares at the ready.
