I would say 12g since some areas (Like around here) will not let you use a rife for hunting. And there are so many options for 12g, from bb shot to 00 buck to slugs.

If it has to be a rifle then
.308

Tough to beat .729 caliber as an all-around firearm, though it is more of a short-range proposition on large game. ;)

30-06 Gets My Vote
 
There was that US military cartridge developed way back in 1903. It was a .30 caliber that was called the 30-03. About three years later they made a couple of improvements to the design, changing the name. After that hunters, shooters and soldiers all around the globe used it to great effect on anything unlucky enough to catch a bullet from one. The new cartridge does well with 125gr to 220gr bullets or so I hear. I can't recall the name given to that 102 year old round but I am sure if you ask nicely one of your fellow High Road members will point you in the right direction.

Deadly accurate, devestating terminal performance and easy to shoot. Other than dangerous, toothy/clawy, charging bruins there is nothing Iwouldn't feel comfortable tackling with it.
 
I'm going to vote for the .338 Win Mag simply for the big Alaskan moose and brown bears. The .300 Weatherby would be good, too.

If handloading was an option, I'd vote for the .358 Norma Mag.
 
What's available locally?

When choosing a caliber I try to consider what's available in the local gun shops where I'm hunting. In the town where I hunt there's a hunting/fishing "sporting goods" shop owned by a local guide. If something happens to the handloads I brought from home or they're left behind (not impossible, my dad once left his license tags home) what is this shop going to carry?

For certain: .30-30, .30-'06, .270 Win, .308 Win, 12 ga.
Probable: .243 Win, .257 Roberts, 7mm Rem Mag, maybe even .25-'06, .35 Rem, and .45-70.
Maybe even some of the Win mag's (.300 Win Mag and .300 WSM come to mind).

So before you bring that 7mm-'08, that .260 Rem, the short action ultra mag, the WSSM, or the foreign surplus rifle. STOP. THINK. Can I find that ammo in the middle of nowhere when the only place that sells ammo is the bait and tackle concession in the local gas station?

BTW my caliber of choice is .30-'06.
 
while the 30/06 is the obvious answer there is a military ctg. tha precieded it by 30 years that has killed a ton of game and continues to be a well respected big game gun today.
I'd take my Marlin 1895CB anywhere.
