357mag (including 38special).

It has tremendous versatility. It can be loaded from superlight plinking loads (factory or handloaded) to heavy bullet hunting loads. It also makes a good SD weapon because it doesn't have to be large and heavy (ultralight scandium guns to large frame steel guns). When you have to buy factory ammo, it can be found relatively cheap. Of the various flavors, a 3-4" midframed 357mag is probably the most flexible gun made. From target to CCW to home defense to small game and maybe large game hunting, it does it all.

45 Colt. If you handload, it will perform just about any task you could expect from a handgun cartridge. 'Sides, there's a lot of history associated with it. :)

Hey Glocker, where's YOUR answer?

For me, .44 Magnum. Same reasons guys gave for the .357, just more of it! :D

.41 magnum
It can do anything I need to do in my neck of the woods.
Deer, possum, raccoons, etc.
I carry a snubby .44 mag as backup, but that's just for a NY reload.

+1 on the .41 mag. Powerful, accurate, easier to control than .44 mag,
and just plain fun. Roll your own for best results.

My first handgun was a .44 Magnum and 30 years later it is still my favorite caliber. :D

Although I shoot several and have tons of fun with them my all time favorite is the .22lr. I burn up more .22lr ammo than anything.

.357 Mag/.38 Special

Because the 3 revolvers that I own are all chambered for that round. Seriously, for me the .357 Mag. meets all my wheel gun needs from target shooting-defensive purposes. I really enjoy shooting the .357 magnum much, much more than the .44 mag.

.357 Mag/.38 Special
I prefer .357 Mag, because it has a reasonable amount of recoil and I feel it is accurate from a variety of barrel lengths. Also it will shoot the .38 round if you are money conscious. I have three of them and carry at least one daily.

.357 magnum, for reasons stated above about shooting the .38's. I think the .357 is the all round best self defense (against badguys, not large game) out their and the selection of wheel guns to shoot them is great. I think .44 magnum would make number 2 on my list.

I think .357mag is the most versatile chambering and it tends to be what I suggest for a first or only revolver for that reason. It can be downloaded with inexpensive and light shooting .38spl target ammo, loaded up with good defensive .38+P, defensive .357mag, or the barnburning hunting magnum rounds. I think .38spl/.357mag in a 2 1/2-4" K-frame or L-frame sized revolver (especially the K-frame size) has to be the most versatile combination of platform and caliber around.

Still, that wasn't the question, it was favorite. It used to be .357mag. All the versatility issues above, and when shooting full-house magnums it is a blast to shoot. However, now it is .41mag. Not as versatile, but so much more fun. It isn't punishing like the .44mag, it is much more powerful than .357mag, and it is a ton of fun.

In my humble opinion, there are four useful handgun cartridges:

1. The .22 LR. No one should be without a .22 LR. They're fun and cheap to shoot, good woods guns, good learning guns, good competition guns.

2. The .357 Magnum -- highly versatile (especially when you include the .38 Special in that category). They'll do anything any lesser centerfire cartridge will do, and do it better.

3. The .45 ACP -- not a revolver round, per se, and best when teamed with the great M1911 for a defensive arm.

4. The .45 Colt -- when the .357 isn't enough.

Most versatile? 44 Magnum - I can download it to cover everything from lite plinking and two-legged defensive work, or upload it and go hunting. I can also find it in virtually any store in the US that sells ammo (unlike 454 Casull or the other bigger/badder chamberings).

My favorite? The 38 Special. It's most often 'just enough' without being too much, and can be chambered in revolvers that don't stretch the budget or the belt too badly.
